Therapy with original anticonvulsant galodif influences on properties of benzodiazepine receptors of thrombocytes and levels of neuroactive steroid hormones in alcoholic patients

Alcoholism is one of forms of impulsive behavior in patients conditioned by common inhibitory deficit. Deeper understanding of mechanisms of regulation of processes of brain excitation, identification of targets for alcohol allows creation of potentially effective therapy directed at removal of consequences of alcohol abuse and withdrawal. Comparative analysis of density of benzodiazepine receptors (BDR) of plasmatic membranes of thrombocytes and levels of neuroactive steroid hormones (NS) has shown a reliable heightening of BDR density, decrease of progesterone (PG) and testosterone (TS) in blood of patients with alcoholism as compared with PG and TS concentration in healthy volunteers. Investigation of level of cortisol (CS) has revealed the considerable heightening of concentration of this steroid in serum of blood of alcoholic patients. Course application of original anticonvulsant Galodif® (meta-chlor-benzhydrylurea) during 21 days at daily dose 300 mg in patients with withdrawal and post-withdrawal state has caused reduction of severity of symptoms, typical for alcohol withdrawal syndrome. Therapy with Galodif® resulted in normalization of balance of NS, modulating function of GABA/BDR, and decrease of density of receptors up to level of control values that is very perspective in treatment of alcohol withdrawal and alcoholism. Galodif® lowered level of CS by 138% and heightened level of PG by 160 % in serum of blood in alcoholic patients approximating them to level of hormones in healthy persons. Alteration of BDR density and levels of steroid hormones in blood of alcoholic patients reflects disturbance of steroid regulation (CS and PG) as one of mechanisms of development of craving for alcohol.
